Antibacterial mechanism of bronopol


Bronopol chemical name is 2-bromo-2-nitro-1,3-propanediol, also known as bromide, bronopol, BNPD, etc., English name is 2-bromo-2-bitro-1,3-propanediol, CAS registration number: 52-51-7, foreign trade name is Bronopol. The structure is as follows:


Pure product is white to light yellow powder or crystal, odorless and tasteless. The temperature of 118 ° C. It has a solubility in water of 25g/100g (25°C); it is insoluble in oil and has a solubility in oil of 0.5g/100g (25°C). The oil/water partition coefficient is small. The aqueous solution slowly decomposes when it is alkaline. Cannot be used with certain metals such as aluminum. This product is unstable under high temperature and alkaline conditions (pH is greater than 8) and turns brown under illumination.

Bronopol is a broad-spectrum fungicide of bromonitroalcohol. It has high bactericidal activity, low concentration and wide range of use. It is non-irritating and non-sensitizing to human skin. It is approved by the United States. One of the cosmetic preservatives used.


Three major antibacterial mechanisms:

(1) The antibacterial activity is derived from the oxidative bromine atom lacking electrons in the molecule. The antibacterial mechanism is that the bromine atom oxidizes the thiol group on the surface of the bacterial cell membrane, making it a disulfide compound, which generates extra large protrusions on the cell wall and causes the cell wall to rupture. The inner dissolved matter flows out to kill the bacteria;


(2) Another possible pathway is the release of activated bromine, which binds to cell membrane proteins to form nitrogen-bromo compounds, which interfere with cellular metabolism and ultimately cause bacterial death;


(3) The formaldehyde released during the decomposition of Bronopol can also solidify the bacterial protein and thus play a bactericidal effect.
